Skin Irritation (ECVAM Validated)
The Model
MatTek's patented EpiDerm¢â System consists of normal, human-derived epidermal keratinocytes (NHEK) which have been cultured to form a multilayered, highly differentiated model of the human epidermis. These "ready-to-use" tissues, also known generically as Normal Human-3D (NHu-3D), are cultured on specially prepared cell culture inserts using serum free medium, attain levels of differentiation on the cutting edge of in vitro skin technology. Ultrastructurally, the EpiDerm Skin Model closely parallels human skin, thus providing a useful in vitro means to assess dermal irritancy and toxicology. The Method

Transfer tissues from agarose to assay medium
-
Incubate (37 ¡¾1¡ÆC, 5 ¡¾1% CO2, 95% RH) for 60 ¡¾5 min
- Transfer tissues to fresh assay medium
-
Incubate (37 ¡¾1¡ÆC, 5 ¡¾1% CO2, 95% RH) overnight (18 ¡¾3 h)
-
Dose 3 tissues each with 30 ¥ìl/25 mg TS, PC, or NC
-
Expose 60 min (37 ¡¾1¡ÆC, 5 ¡¾1% CO2, 95% RH)
-
Stop exposure after 60 minutes by rinsing in DPBS
-
Transfer tissues to fresh assay medium
-
Incubate (37 ¡¾1¡ÆC, 5 ¡¾1% CO2, 95% RH) for 24 ¡¾2 hours
-
Collect medium for IL-1¥á analysis (optional)
-
Incubate (37 ¡¾1¡ÆC, 5 ¡¾1% CO2, 95 % RH) for 18 ¡¾2 hours
-
Blot tissues and perform MTT assay

Read OD in a plate spectrophotometer at 550-570nm
